GS-441524 exerts its antiviral effect by acting as a chain terminator during viral RNA synthesis. After cellular uptake, it is phosphorylated into its active triphosphate form (GS-443902), which mimics adenosine triphosphate. This active metabolite competitively inhibits the viral RNA-dependent RNA polymerase (RdRp) and, when incorporated into the growing RNA strand, causes premature chain termination, effectively halting the replication of RNA viruses such as coronaviruses and filoviruses.
