1.MECHANISM OF ACTION
This peptide functions as a specific antigen for CD8+ cytotoxic T lymphocytes (CTLs) in murine systems. In GPC3-overexpressing mouse tumor cells, intracellular processing generates this peptide fragment, which is then presented on the cell surface bound to a specific MHC class I molecule (e.g., H-2Kᵇ). As an exogenous synthetic peptide, it can be loaded onto the same MHC class I molecules on professional antigen-presenting cells (APCs) in vitroor in vivo. The peptide-MHC complex is specifically recognized by T-cell receptors (TCRs) on GPC3 (298-306)-specific CD8+ T cells.
2.PRODUCT INDICATIONS
Mouse Model Development: Inducing and studying GPC3-specific T-cell responses in syngeneic mouse models of hepatocellular carcinoma (HCC), hepatoblastoma, or melanoma that express murine GPC3.
In VivoImmunotherapy Studies: Evaluating the efficacy of peptide-based vaccines, dendritic cell vaccines, or adoptive T-cell therapies targeting GPC3 in immunocompetent mice.
In VitroT-Cell Activation: Stimulating and expanding GPC3-specific CD8+ T-cell lines or clones from the splenocytes of immunized or tumor-bearing mice for functional assays (cytotoxicity, cytokine release).
Immune Monitoring in Mice: Detecting and quantifying GPC3-specific T-cell responses in murine blood, spleen, or tumor tissue using techniques such as interferon-gamma ELISPOT, intracellular cytokine staining (ICS), or MHC tetramer staining.
3.CLINICAL EFFICACY OF PRODUCT
Preclinical Proof-of-Concept: Studies using this and similar murine epitopes have demonstrated that GPC3-targeted vaccines and adoptive T-cell therapies can induce potent antitumor immunity and inhibit tumor growth in mouse models of liver cancer. This validates GPC3 as a promising immunotherapy target.
Translational Bridge: Successful immunotherapy targeting this murine epitope provides the rationale and preclinical data necessary to develop analogous therapies for humans. Human GPC3 is also an oncofetal antigen overexpressed in HCC, making it a prime target.
Human Clinical Context: Based on strong preclinical data, multiple human GPC3-targeted therapies are in clinical development, including:
Peptide Vaccines: Targeting human GPC3 epitopes in patients with HCC.
CAR-T Cell Therapies: Chimeric antigen receptor T-cells targeting GPC3.
Bispecific Antibodies: Redirecting T-cells to GPC3+ tumors.
Status: This specific mouse peptide is not a therapeutic. Its "efficacy" lies in its ability to generate robust preclinical data that de-risks and informs the development of human GPC3-directed immunotherapies currently in clinical trials.
