LH-RH exerts its effects by binding and activating the specific Gonadotropin-Releasing Hormone Receptor (GnRHR) on the surface of pituitary gonadotrope cells. GnRHR is a G-protein coupled receptor (GPCR) primarily coupled to the Gq/11 protein. Binding triggers phospholipase C (PLC) activation, leading to the hydrolysis of phosphatidylinositol 4,5-bisphosphate (PIP₂) into inositol trisphosphate (IP₃) and diacylglycerol (DAG). IP₃ mobilizes intracellular calcium, while DAG activates protein kinase C (PKC).
