This substrate operates on a protease cleavage-release mechanism. The peptide moiety (Juice-WATER) is specifically recognized and cleaved by active proteases present in the juice or enzyme preparation. Upon cleavage, the para-nitroaniline (pNA) group is liberated from the peptide backbone. Free pNA absorbs light strongly at a wavelength of 405-410 nm, producing a yellow color. The rate of increase in absorbance at this wavelength is directly proportional to the protease activity in the sample, allowing for precise kinetic or endpoint analysis.
