1.MECHANISM OF ACTION
Calcitonin exerts its effects by binding to the Calcitonin Receptor (CTR), a G protein-coupled receptor, primarily on osteoclasts and renal tubular cells:
Inhibition of Bone Resorption: In bone, it binds to receptors on osteoclasts, the cells responsible for breaking down bone. This binding inhibits osteoclast activity and motility, reducing bone resorption and the release of calcium and phosphate into the blood.
Enhancement of Renal Calcium Excretion: In the kidneys, it acts on the distal tubules to inhibit calcium and phosphate reabsorption, increasing their excretion in the urine.
Analgesic Effect: Calcitonin has a direct analgesic effect on the central nervous system, independent of its effects on calcium, which is beneficial in painful bone diseases like Paget's disease and osteoporosis.
