This substrate operates on a protease cleavage-release principle. It consists of a short peptide sequence (commonly DEVD) that mimics the native cleavage site of caspase-3, linked to a chromogenic reporter molecule, most commonly para-nitroaniline (pNA). During apoptosis, active caspase-3 specifically recognizes and cleaves the peptide bond after the aspartic acid (D) residue in the DEVD sequence. This cleavage liberates the free pNA chromophore, which absorbs light strongly at a wavelength of 405-410 nm, producing a yellow color. The rate of increase in absorbance is directly proportional to the amount of active caspase-3 in the sample.
The Caspase-3 Substrate itself is a research tool, not a therapeutic. Its clinical and translational efficacy lies in its vital role as a standardized biomarker assay. It is used extensively in: 1) Preclinical Drug Development: To evaluate the efficacy of novel anti-cancer drugs by confirming they induce apoptosis in target cells. 2) Toxicology Studies: To assess the apoptotic potential of new chemical entities. 3) Basic Cancer Research: To understand dysregulated apoptosis in tumors and resistance mechanisms. The data generated using this substrate directly informs the progression of potential therapies into clinical trials and helps elucidate disease mechanisms.
