1.MECHANISM OF ACTION

This substrate operates on a protease cleavage-release principle. It consists of a short peptide sequence (commonly DEVD) that mimics the native cleavage site of caspase-3, linked to a chromogenic reporter molecule, most commonly para-nitroaniline (pNA). During apoptosis, active caspase-3 specifically recognizes and cleaves the peptide bond after the aspartic acid (D) residue in the DEVD sequence. This cleavage liberates the free pNA chromophore, which absorbs light strongly at a wavelength of 405-410 nm, producing a yellow color. The rate of increase in absorbance is directly proportional to the amount of active caspase-3 in the sample.
