1.MECHANISM OF ACTION
This peptide functions as a specific T-cell receptor (TCR) antigen in the context of cancer immunology. Intracellularly, it is processed from the full-length gp100 protein and presented on the surface of melanoma cells by the HLA-A0201 molecule. As an exogenous synthetic peptide, it can be loaded onto the HLA-A0201 molecules of antigen-presenting cells (APCs) in vitro. When presented, the peptide-MHC complex is recognized by the complementary TCRs on CD8+ cytotoxic T lymphocytes (CTLs).
2.PRODUCT INDICATIO
Melanoma Immunology Research: Studying antigen-specific T-cell responses, immune evasion mechanisms, and epitope spreading in the context of gp100-expressing melanomas.
In VitroCTL Activation & Expansion: Stimulating and expanding gp100 (154-162)-specific human CD8+ T-cell cultures from peripheral blood mononuclear cells (PBMCs) of HLA-A*0201+ donors for functional assays.
Immune Monitoring Assays: Serving as the target antigen in enzyme-linked immunospot (ELISPOT) assays, intracellular cytokine staining (ICS), or tetramer staining to detect and quantify antigen-specific T-cell responses in patient samples or vaccine trials.
Cancer Vaccine Research: Evaluating this epitope as a component in peptide-based, dendritic cell-based, or other vaccine strategies against melanoma in preclinical models.
TCR Affinity & Specificity Studies: Used in surface plasmon resonance (SPR) or tetramer dissociation assays to measure the binding kinetics of T-cell receptors specific for this epitope.
3.CLINICAL EFFICACY OF PRODUCT
Research & Preclinical Validation: This epitope is one of the most extensively studied melanoma antigens. It has been shown to elicit potent and specific CTL responses in vitroand in animal models, validating its immunogenicity.
Clinical Trial History: Peptide vaccines containing this epitope, often in modified forms for higher HLA binding (e.g., gp100:209-217(210M)), have been tested in numerous clinical trials for melanoma, often in combination with high-dose interleukin-2 (IL-2).
Therapeutic Context: While peptide vaccines alone showed modest clinical activity, this epitope remains a critical target for adoptive cell therapy (ACT). In ACT, patient T-cells are genetically engineered to express TCRs specific for this gp100 epitope, leading to robust tumor regression in some clinical trials.
Current Status: The gp100 (154-162) epitope is not an approved standalone drug. However, it is a prototypical "shared tumor antigen" that continues to be a benchmark for developing and testing novel T-cell-based immunotherapies. Its efficacy is realized through engineered cellular products, not as a direct therapeutic peptide.
