GIP, Porcine (Gastric Inhibitory Polypeptide), also known as Glucose-dependent Insulinotropic Polypeptide, is a 42-amino acid incretin hormone isolated from or synthesized to match the porcine (pig) species. This peptide is secreted by K-cells in the duodenum and jejunum in response to nutrient ingestion. While named for its initial discovery as a gastric acid inhibitor, its primary and more significant role is as a potent insulinotropic hormone.

Porcine GIP exerts its effects primarily by binding and activating the GIP Receptor (GIPR), a G-protein coupled receptor (GPCR) expressed on pancreatic beta cells, adipocytes, osteoblasts, and other tissues. Activation of GIPR is strongly glucose-dependent. In the pancreas, receptor binding stimulates Gs-protein-mediated activation of adenylate cyclase, leading to a sharp increase in intracellular cyclic AMP (cAMP).
