Within melanoma cells, the intracellular proteasome processes the full-length gp100 protein, generating this 9-mer fragment, which is then transported into the endoplasmic reticulum, loaded onto HLA-A0201 molecules, and presented on the cell surface. As an exogenous synthetic peptide, it can be directly loaded onto the HLA-A0201 molecules of antigen-presenting cells (APCs) in vitro. The resulting peptide-MHC complex is specifically recognized by T-cell receptors (TCRs) on gp100 (639-647)-specific CTLs.
