Histatin 5 is a natural, histidine-rich, cationic antimicrobial peptide consisting of 24 amino acids (DSHAKRHHGYKRKFHEKHHSHRGY). It is one of the major functional peptides derived from the Histatin family, primarily secreted by the parotid and submandibular glands in human saliva. Histatin 5 is renowned for its potent, broad-spectrum antifungal activity, particularly against Candida albicans, and exhibits bactericidal properties. It also plays roles in wound healing and enamel pellicle formation.

1.MECHANISM OF ACTION

Histatin 5 exerts its primary antimicrobial effects through a multi-step mechanism culminating in membrane disruption and intracellular targeting. Initially, the cationic peptide binds to negatively charged components on the microbial cell surface (e.g., phospholipids in fungal membranes or lipopolysaccharides in bacteria). This electrostatic interaction is followed by peptide translocation across the membrane without massive pore formation, a process distinct from many pore-forming AMPs. Once inside the fungal cell, Histatin 5 targets mitochondria, where it induces non-lytic efflux of cellular ATP and other small molecules, disrupts mitochondrial membrane potential, and triggers the generation of reactive oxygen species (ROS).
