Neuromedin B, porcine is an endogenous bombesin-like peptide and a high-affinity ligand for the neuromedin B receptor (NMB-R), a G-protein coupled receptor (GPCR) widely expressed in the central nervous system (CNS), gastrointestinal tract, respiratory system, and certain tumor cells. With an IC₅₀ value of 2 nM for NMB-R binding, it activates Gq/11 and Gi/o-dependent intracellular signaling pathways upon receptor ligation. This activation triggers hydrolysis of phosphatidylinositol 4,5-bisphosphate (PIP2) to generate inositol trisphosphate (IP3) and diacylglycerol (DAG), elevating intracellular calcium levels to mediate smooth muscle contraction
