[Ser2]-Neuromedin C is a modified bombesin-like peptide, featuring a serine substitution at position 2 of native Neuromedin C, that acts as a selective ligand for bombesin receptor subtype 2 (BB₂R), a G-protein coupled receptor (GPCR) widely expressed in the gastrointestinal tract, pancreas, central nervous system, and BB₂R-overexpressing tumor cells. Upon binding to BB₂R, it activates Gq/11-dependent intracellular signaling pathways, triggering hydrolysis of phosphatidylinositol 4,5-bisphosphate (PIP2) to generate inositol trisphosphate (IP3) and diacylglycerol (DAG). This cascade elevates intracellular calcium levels, inducing smooth muscle contraction
