Tyr-Uroguanylin (rat) functions as a guanylate cyclase-C (GC-C) receptor agonist, binding to the extracellular domain of GC-C receptors expressed on intestinal and renal epithelial cells. This binding activates the intracellular catalytic domain, leading to the conversion of GTP to cyclic GMP (cGMP). The elevated intracellular cGMP levels activate protein kinase G (PKG), which phosphorylates downstream effectors including the cystic fibrosis transmembrane conductance regulator (CFTR) chloride channel. This signaling cascade stimulates chloride and bicarbonate secretion while inhibiting sodium absorption, ultimately regulating fluid and electrolyte transport. The N-terminal tyrosine modification does not significantly alter the peptide's receptor-binding affinity or biological activity, allowing it to function similarly to native uroguanylin while providing additional utility for detection and labeling applications.
